Planting Test of Three Varieties of Papaya in Vitro Plantlet at Yuanmou, Yunnan

Abstract:
By determining growth properties and investigating diseases and insect pests of three varieties of papaya in vitro plantlet (qingyou,qingpi and solo), which were planted at Yuanmou, Yunnan, some important data were obtained. These results not only validate the environmental suitability and disease resistance of papaya in vitro plantlet effectively, but also provide the theoretical information for expanding cultivating of papaya in vitro plantlet on a large scale and papaya breeding for disease resistance.
Cloning of Ascorbate Peroxidase Gene HbAPX in Hevea brasiliensis and Its Response to Salicylic Acid

Abstract:
Based on the sequencing results of RNA-Seq analysis of Hevea brasiliensis and the amino acid sequence of APX1 in Arabidopsis thaliana, the specific primers were designed and RT-PCR were performed to amplify the APX cDNA of Hevea brasiliensis(HbAPX). The results showed that the open reading frame of HbAPX is 858bp encoding 285 amino acids with a conserved ascorbate peroxidase domain; the amino acid sequence of the HbAPX has 90% homology with APX from Jatropha curcas, and the homology is more than 80% with APX from Populus tomentosa, Gossypium hirsutum, Arabidopsis thaliana, Oryza sativa; the phylogenetic analysis data indicated that HbAPX belong to peroxisome type APX; the quantitative analysis data suggested that the expression of HbAPX was induced by SA, which is probably involved in system acquired resistance and hypersensitivity reaction in rubber.
Expression of Porphyromonas Gingivalis Fimbrillin Gene in the Fruit of Transgenic Tomato and Its Immunization Assay

Abstract:
In this report, Agrobacterium-mediated transformation of tomato with plasmid pBIE8-CTB-FimA(266-337) containing FimA and CTB gene with E8 promoter were performed, and the immunity of the transgenic tomato fruit as an edible vaccine by oral immunization of mice were determined. The results indicated that the transgenic plants were confirmed by Southern blot and RT-PCR; the fusion proteins from leaf and fruit tissue of transgenic plants were detected by Western blot; specific anti-FimA antibodies in the serum of mouse were detected, all of which suggested that candidate edible plant vaccine was obtained successfully.
Study on the Spawn Isolation of Wild Anoderma Calidophilum

Abstract:
The surface sterilization of fruit body was performed via a method which was similar to the surface sterilization for the explants in plant tissue culture to treat wild Anoderma calidophilum, and the little pieces of the fruit body were cut down and used for the in vitro culture, and the "small-container-cultivation" method was applied to determine the isolation and purification results of spawn. The result showed that the method for the spawn isolation of G.calidophilum was rapid, simple and effective, by which purified spawn were successfully isolated from 9 fruit bodies of G.calidophilum, respectively.
Interspecies Competition between Tetraychus urticae Koch and T.cinnabarinus Boisduval Fed with Cotton

Abstract:
Based on the comparing of population dynamics of single species and mixed population and the intrinsic rate of increase(rm) when Tetraychus urticae Koch and T.cinnabarinus Boisduval fed with cotton, the interspecies competition were studied. The results showed that it was 38th day when the amounts of active mites and mature mites of T.urticae and T.cinnabarinus reach to peak in the single population; and it is 48th and 54th day when the amounts of active mites and mature mites of T.urticae or T.cinnabarinus reach to peak in the mixed population, and the amounts of Tetraychus urticae Koch are 1 times higher than that of T.cinnabarinus Boisduval. Both in the single population and mixed population,the intrinsic rm of T.cinnabarinus active mites and mature mites was higher than that of T.urticae, which suggested interspecies competition of T.cinnabarinus was higher than T.urticae fed on cotton.
Morphological Variation Analysis of Five Different Geographic Populations of Paphia textile Along South China Sea

Abstract:
The multivariation analysis methods(cluster analysis, principal component analysis and discriminate analysis) were used to compare the eleven morphological characters of five populations of Paphia textile in Zhanjiang, Shenzhen, Shantou, Sanya, and Xiamen. The results of clusters analysis and principal component analysis indicated that the morphological characters were similar among the populations in Sanya and Xiamen, while the polulations in Zhanjiang, Shenzhen and Shantou were morphologically grouped together, all of which were apparently divided into two groups. Three principal components were constructed by the principal component analysis method, the contributory ratio of the first principal component was 37.489%; that of the second principal and the third principal were 17.803% and 10.704%, respectively; the cumulative contributory ratio was 66.005%.Then, the discrimination functions of the five populations were set up, the identification accuracy was 43.3%-66.7% (P1) and 43.5%-72.7%(P2). The total discriminate accuracy was 52.6%. These data suggested that the morphological variations of Paphia textile were resulted from the joint effects of genetic components and environment, Combined with studies of molecular genetics, the genetic characteristics of the five populations will be described more exactly, which can provide science basis for germplasm resources protection and genetic breeding.
Purification and Characteristic of Albumin from Monopterus albus Skin

Abstract:
The anion exchange chromatography and gel filtration chromatography were performed and skin albumin of Monopterus albus was obtained. The protein is a single chain protein with molecular weigh of 67×103, and named MAA-skin. MAA-skin was determined as a glycoprotein by Sciff’s agent. The N-terminal amino acid sequence is GHVKW. The purified MAA-serum was used to immunize rabbits to prepare polyclonal antibody. ELISA data demonstrated that the antibody titer of the MAA-skin was up to 1∶ 10 000. Western blot results indicated that the polyclonal antibodies raised against MAA-serum strongly cross-reacted with both MAA-serum and MAA-skin with a compatible grade. The distribution of frog albumin in frog skin suggested that it plays important roles in skin physiological functions, such as water salt metabolism, osmotic pressure maintaining and substance transporting, etc.
Studies on Influenc ing Factors of Cassava Regeneration System

Abstract:
In this report, Varieties of SC6, SC7, SC8, SC124, NZ188, C3 and C4 were used as experimental materials, several significant factors which influence the regeneration system of cassava were studied. The results indicated that 6-BA had a significant inhibitory effect on apical dominance of cassava, the higher the concentration was, the stronger the inhibition effects was; 2,4-D and picloram had almost equal effects for promoting somatic embryogenesis, in the process of induction of primary somatic embryos, embryogenesis rate of all breeds reached 100%, except SC7 remained a low embryogenesis rate of secondary somatic embryos, embryogenesis rate of secondary somatic embryos of other varieties went up to 90%; the ripening time of somatic embryos had a great influence on the organogenesis of cotyledon, frequency of cotyledon organogenesis was higher around 2 weeks after embryo maturation and the induction frequency could be more than 70%; the somatic embryogenesis ratio and cotyledon organogenesis frequencies were very different among distinct genotypes,SC8 and NZ188 cultivar had the highest and second-highest frequency respectively,SC7 cultivar displayed the lowest frequency; The addition of AgNO3(5 mg·L-1) into subculture could help improving the organogenesis capacity of mature cotyledonary.
Effects of Different Treatment Methods on Seed Germination of Tadehagi triquetrum(L.) Ohashi

Abstract:
In this report, the seeds of Tadehagi triquetrum(L.) Ohashi were treated with different methods, and the germination potential and germination rate of the treated seeds were statistically analyzed. The results indicated that the germination potential and germination rate of the seeds treated with 98% H2SO4 for 10 minutes was 70% and 81% respectively, which was the highest. The method is easy to operate,low-cost, and by which the germination of seeds were faster. The method was recommended for using in actual production.
Karyotype Analysis of Ten Cucumis melo Germplasms

Abstract:
The root tips of germinating seeds were used as materials and chromosome counting and karyotype analysis were performed in eleven melon relative plants and one wild melon plant. The results showed that chromosome number in somatic cells of most samples were 2n=24, except one accession C.anguria with 48 chromosomes and one accession C.dipsaceus with 44 chromosomes in somatic cells; The karyotypes were divided into five types: C.bisexualis is 2n =2x=2 M+14 m +10 sm,C.hystrix is 2n =2x=24 =24 m,C.dipsaceus and C.prophetarum 2n =2x=20 m +4 sm,C.myriocarpus, C.ficifolius, C.africanus and C.pustulatus is 2n =2x=12 m +12 sm,C.zeyheri is 2n =2x=24 =2 M +18 m +4 sm, and there were no satellites detected in hemorphological observation of chromosomes.
Effects of Different Packaging Material and Preservation Thickness on Storage Characteristic of Fresh-cut Melon

Abstract:
In order to investigate the effects of packaging material and preservation thickness on the storage characteristic of fresh-cut melon, monolayer and nondegradable film, bilayer and nondegradable film, monolayer and degradable film, bilayer and degradable packaging materials were used for the package of fresh-cut melon. The results showed that bilayer film packaging delayed the deterioration, reduced weight loss effectively and maintained the appearance quality better; bilayer and degradable film packaging maintained the high content of VC and activities of POD and SOD. The conclusions were drawn that bilayer and degradable film is a good material used for the storage and preservation of fresh cut melon.
Effects of Low Temperature on Expression of Genes Encoding Entrance Enzyme of Grape Shikimate Pathway and Key Enzymes of Post Chorimate Pathway

Abstract:
In this report, grape (Vitisvinifera L. cv. ‘Cabernet Sauvignon’) berries at 3, 7 and 11 weeks after full bloom were used as materials, real time PCR were performed, and the effects of treatments with constant low temperature and variable low temperature, respectively, on the expression of the genes encoding 3-Deoxy-D-arabino-heptulosonate-7-phosphate synthase (DAHPS), an entry enzyme of shikimate pathway, and anthranilic acid synthase (AS) and chorismatemutase (CM), key enzymes of post-chorismate pathway were investigated. The results showed that the effects were developmental stage dependent, the treatments with constant and variable low temperatures, both significantly increased the transcript abundance of three VvDAHPS isogenes and VvCM-1 in 3-week berries; the activation extent of expression of VvDAHPS-1 and VvDAHPS-2 was much higher than that of VvDAHPS-3 and VvCM-1, which were greatly reduced accompanying with berry mature; the expression of VvCM-2 and VvAS was slightly affected by two low temperature treatments. Additionally, the response of these tested genes to low-temperature treatments was time dependent. These data suggested that a certain low-temperature will stimulate carbon flow in young grape berries to shikimate pathway, leading to the production of more secondary metabolites.
Effects of 6-BA and NAA on Immature Embryo of Areca catechu L. Cultured in vitro

Abstract:
The immature embryo of "Reyan No.1" Areca catechu L. was used as explants in vitro, and the effects of different concentration of 6-BA and NAA on the embryo germination and growth were compared. The results showed that when the concentration of 6-BA was 2 mg·L-1, the growth of embryo was the fastest, and the development of bud and root were regular; when the concentration of NAA was 0.5 mg·L-1, the growth of root was the best, and the development of young seedlings were regular. These data provided important technical means for the tissue culture, resource conservation and utilization of arecanut.

Effects of Different Shading Rate on the Growth and Quality of Oncidium Cut Flower

Abstract:
In this report, the effects of different adjustment shading rates on Oncidium cut flower quality were compared. The results showed that it is under the circumstance of 75% shading rate which is good for the growth and development of Oncidium seedlings; when Oncidium buds grow up to 10-20 cm, 60% shading rate is necessary, which can greatly up-righten and lessen lodging heads of the flowers, shorten Oncidium cut flower panel length, reduce under branch height and improve cut flower quality, but has no significant effect on the numbers of branch.
Study on Preparing SiO2/NR Composite with Water Glass

Abstract:
In this report, SiO2 sol gel prepared with the cheap water glass was mixed and coagulated with nature rubber latex to prepare the SiO2/NR composite. The properties of composites were investigated by TG and DMA. The results showed that dispersion of SiO2 solgel particles in NR matrix increased and interface compatability between NR and SiO2 improved. The thermal stability and mechanical properties of composites were improved.

Research Progresses on Triacylglycerol Lipase

Abstract:
In this paper, the research progresses of TGL were reviewed. Triacylglycerol lipase(TGL)which is highly conserved in evolution mainly involved in TAG hydrolysis; the analysis results of the protein structure of TGLs showed that the proteins have patatin domain containing conserved motif GXSXG; TGL are usually located in microbial oil droplets, plant seeds and animal’s adipose tissue, however, its mRNAs were also detected in plant chloroplasts and other animals tissues such as cytoplasm; the lipase activities of TGLs were very high, which contribute to various metabolic processes which have relations with obesity, diabetes, and cardiovascular diseases, additionally, TGL also play important roles in various developmental stages of plants such as seed germination, seedling growth and leaf senescence.
