Influences of Metabolites of Pinctada martensii on the Growth of Sargassum fusiforme

Abstract:
During the polyculture of shellfish Sargassum fusiforme and algae Pinctada martensii,we compared the growth differences of algae in the experimental group with shellfish,the simulation group without shellfish and the control group by measuring the amount of nitrogen and phosphorus released by the shellfish and absorbed by the algae.Furthermore,the removal rate and the absorptivity of the algae for the metabolic products of the shellfish were also determined.Results showed that NH+4-N is the main form of nitrogen which P.martensii released and S.fusiforme absorbed.Four metabolites are NH+4-N,NO-3-N,NO-2-N and PO43+-P,and their release ratio from the shellfish is 27:21:1:18 while the algae absorption ratio 17:12:1:2.6.Analysis of variance showed that algae in the experimental group played a significant role in promoting the absorption rate of nitrogen and phosphorus(P<0.05) when compared to the simulation group.Meanwhile,Duncan’s multiple comparisons between the groups showed that daily weight gain and daily growth gain of S.fusiforme were significantly different among all the groups(P<0.05) and increased in the order of the experimental group>the control group C>the stimulation group>the control group A>the control group B,which indicates that the metabolites of P.martensii can promote the growth of S.fusiforme.This research provides a theoretical basis for optimizing the polyculture density of P.martensii and S.fusiforme.
Effects of Different Temperatures and Salinities and pH Values on the Early Development of Plectropomus leopardus Lacépède

Abstract:
The incubation and hatch periods,hatchability and deformity rates of fertilized eggs,and the survival activity index(SAI) of newly hatched larvae of Plectropomus leopardus Lacépède were observed at the temperatures of 20,22,24,26,28,30 and 32℃,salinities of 4.0,9.0,14.0,19.0,24.0,29.0,34.0 and 39.0,and pH values of 5.0,5.5,6.0,6.5,7.0,7.5,8.0,8.5,9.0,9.5.The result showed that the fertilized eggs of P.leopardus were hatched at the temperature range between 24℃ and 30℃ with the optimum temperature being 26~28℃,at the salinity between 25 and 45 with the optimum salinity being 25~30,and at the pH values of being 6.0~8.5 with the optimum pH values being 7.0~8.5.The survival activity index showed that larvae had a fit survival temperature range of 20 ~30℃ with the optimum temperature range between 20 and 26℃,a fit salinity range of 10~40 with the optimum salinity of 20~35,a fit pH value range of 5.5~8.5 with the optimum pH values of 7.0~8.5.The fertilized eggs and larvae of P.leopardus Lacépède shared similar tolerance to the three different environmental factors,temperature,salinity and pH.
Cloning and Expression Analysis of Gene Encoding Shikimate Kinase in Grapes

Abstract:
A shikimate kinase gene named VvSK was cloned from grape(Vitis vinifera L.Cabernet Sauvignon) berries by means of both in silico cloning and molecular cloning.The full-length cDNA of VvSK contains an open reading frame of 906 bp which encodes a polypeptide of 301 amino acid residues with a molecular mass of 33.2 kD and a pI value of 8.6.The VvSK gene from genomic DNA located on the 7th chromosome consists of 4 309 bp,with 10 exons and 9 introns.The sequence homology comparison revealed that the VvSK sequence exhibits a high homology(upto 64.52%) at amino acid level with other plant SK proteins from the GenBank.Phylogenetic analysis indicated that the VvSK has a close evolutionary relationship with Populus trichocarpa SK.The analysis of real-time PCR showed the transcriptional expression of VvSK was detected in all the tested tissues including fruit,stem,leaf and petiole.But some differences in transcript abundance were detected in the skin,pulp and seeds at the different fruit development stages.UV-C treatment induced higher expression of VvSK in the 3-week and the 11-week berries than other treatments(UV-A and UV-B).
Cloning and Functional Analysis of Promoter AtNUDT2 in Arabidopsis thaliana

Abstract:
5’-UTR of AtNUDT2 was cloned by PCR from Arabidopsis thaliana and inserted into pBAR-GUS to form a recombined construct pNUDT2P-GUS.The construct was then used to transform wide-type A.thaliana through Agrobacterium tumefaciens by mediating.A batch of transgenic A.thaliana with 5’-UTR of AtNUDT2 was obtained through glufosinate screening.The transgenic plants(2 weeks old) were then histochemically stained to assess their GUS activity,and the leaves of the 3.5~4 weeks old plants were inoculated with pathogens to observe their pathogenic expression.The results showed the promoter of AtNUDT2 produced is constitutive.Furthermore,the expression of the promoter were not induced by the Pst.DC3000 and Pst.DC3000 AvrB.
Construction and Autoactivation Test of a Bait Vector of AtGRP7 for Two-hybrid System

Abstract:
AtGRP7 is one of slave oscillator of Arabidopsis thaliana,but its physiological role is so far less known.In order to screen the AtGRP7 interacted proteins,the CDS sequence of AtGRP7 was first amplified using PCR and inserted into pGBKT7 vector between the clone site NcoⅠ and PstⅠ to construct the bait vector for yeast two-hybrid(Y2H) system.Restricted digestion and sequencing result showed that the AtGRP7 sequence was inserted into pGBKT7 vector in correct reading frame.Furthermore,the bait vector was transformed into Y2H Gold yeast strain to test the autoactivation and toxicity.The transformed yeast grew very well on SD/-Trp plate,but weakly on SD/-Trp/-Ade plate.No yeasts grew on SD/-Trp/-His and SD/-Trp/-Ade/-His plates,suggesting that histidine synthase gene was not transcribed and translated.This indicated that AtGRP7 was not autoactivated in the Y2H system.
The Construction of Mutant eIF4E in Papaya

Abstract:
Mutations in or loss of the eukaryotic translation initiation factor 4E(eIF4E) can affect infections of viruses to plants,which leads to resistance of the plants against the viruses.The interaction between PRSV-Vpg and Cp-eIF4EeIF4E in Papaya) has been verified.In this study,six point mutations were determined by protein sequence alignment and analysis of homology modeling,and the point mutations were produced in Cp-eIF4E by overlap extension PCR.Then,the mutant Cp-eIF4E genes were connected into two sets of expressive vectors,yeast two-hybrid system and bimolecular fluorescence complementation,which lays a foundation for the follow-up verification of interaction and resistance in the plants.
Test of Tumor-inhibition of Benzyl Isothiocyanate(BITC)in Papaya Seed

Abstract:
Benzyl isothiocyanate(BITC) was extracted from papaya seeds(Carica papaya L.) and used to determine its inhibitory effect on the cell growth of 6 common cancers in vitro including liver cancer by using the MTT method.The results showed the papaya seed extract with BITC at the concentration of up to 5 μmol.L-1(equivalent to 0.745 mg.L-1) inhibited more than 70% of the cell growth of a human liver cancer cell line HepG2,a lung cancer cell line A549,a breast cancer cell line MCF-7,a colon cancer cell line HCT-8,a cervical cancer cell line Hela and a prostate cancer cell line DU-145.Of them the HepG2 liver cancer cells were the most sensitive to BITC which gave an 87% inhibition over the cells at the concentration 2.5 μmol.L-1.HCT-8 colon cancer cells had a similar inhibition rate when treated with BITC at both 5.0 and 10 μmol.L-1.This showed that higher concentration of BITC in the extract of papaya seeds was similar to the lower concentration(5.0 μmol.L-1) in inhibition of the growth of the cancer cells.
Chromosome Observation Method for Cassava Tender Leaves

Abstract:
Modified phenol fuchsin squashing method was used to prepare chromosome specimens of tender leaves of polypoidy cassava SC 124.The tender leaves were pretreated with 0.002 mol.L-1 8-hydroxyquinoline at three different temperatures for different time by using different solution methods and sampled from different parts of the plant.The tender leaves collected 1.5-2 cm long in the morning at 9:00 am-10:00 am were pretreated with 0.002 mol.L-1 the 8-hydroxyquinoline at 4 C for 4 h,and then put in 1 mol.L-1 hydrochloric acid solution at 25 C for 1 h for maceration,and the mesophyll in the central part of the tender leaves were stained with modified phenol suchsin and then squashed for observation.This method produced a well dispersed,clear chromosome image.
Cloning and Sequence Analysis of fgb2 Gene in Fusarium oxysporum f.sp.cubense

Abstract:
It is very important to know the roles of fgb2 genes in invasion process of Fusarium oxysporum f.sp.cubense(FOC) into banana and its relationship in pathogenic difference between the races 1 and 4 of FOC attacking the same banana cultivar.The fgb2 genes were amplified by using Polymerase Chain Reaction and Reverse Transcription Polymerase Chain Reaction,and the amplified products were then sequenced and aligned with DNAStar Meglignment software.The functions of the proteins encoded by fgb2 genes were also analyzed.The results revealed that the open reading frames of fgb2 genes from races of FOC were 1 466 bp and 1 452 bp,encoding 299 and 316 amino acids.FOC race 1 showed a 96.33% similarity of fgb2 gene to FOC race 4.The sequences of fgb2 genes and amino acid sequences of the two FOC races showed no relationship between the pathogenic difference and the fgb2 gene.These results would help to understand the function of fgb2 genes in followup research activities.
Colonization of Trichoderma H6 in Banana and its Effect on the Growth of Banana

Abstract:
Trichoderma H6 was tagged with antibiotic resistance,and inoculated into the potted banana plants to observe its colonization in the plants and its effect on the plant growth.The results showed that Trichoderma H6 was conducted and colonized in the roots,corms,pseudostems and leaves of the banana plants,and that the banana plants colonized in vivo with Trichoderma H6 grew obviously better than the control plants without in vivo colonization of Trichoderma H6,indicating Trichoderma H6 would improve the growth of banana plants when colonized in the banana plants.
Preparation and Bioassay of Harpin Nanoparticles

Abstract:
HarpinXoo and HarpinXm are derived from Xanthomonas oryzae pv.oryzae and Xanthomonas citri subsp.malvacearum,respectively,and have the characteristics of Harpin.HarpinXoo and HarpinXm were prepared as nanoparticle agents.Both the two proteins and their protein nanoparticles induced hypersensitive response and oxidative burst in tobacco leaves,playing a beneficial role in the process of tobacco seed growing,especially HarpinXm nanoparticles which promoted oxidative burst of tobacco by 104.5%.HarpinXoo and HarpinXm nanoparticles showed a burst release at the initial stage,and their release rates increased to 83.9% and 84.5%,respectively,within 48 h.These two nanoparticles played a role in promoting protein activity continuously.
Effect of KGM Compound Film on Post-harvest Storage of Mango Fruit at Ambient Temperature

Abstract:
Fresh fruit of mango variety Haden 1(Mangifera indicus L.) were packaged with KGM compound film and polyethylene film with no film as the control,and stored at ambient temperature(RH=50%,T=25℃).The mangoes treated were determined during the shelf life in terms of hardness,disease incidence,bad fruit rate,weight loss rate,soluble solid content,respiration rate,Vitamin C content,titratable acid and SOD,PPO and POD activities.Results indicated that the KGM compound film and polyethylene film exhibited the highest respiration peak on the 12th day,6 days late as against the control,and a higher SOD content at the day 9,1.75 times higher than the control.The KGM compound film and polyethylene film also significantly reduced mango disease incidences,bad fruit rate and water loss rate,and had higher fruit hardness and Vitamin C content but lower PPO and POD activities in the peel,effectively restraining occurrence of peel browning during mango storage,accumulation of soluble sugar and degradation of titratable acid,and retarding mango ripening.KGM compound film gave a better effect than polyethylene film in improving the mango quality during the shelf life.
Effect of Coatings on Storage of Pomelo

Abstract:
Konjac glucomannan complex film solution,carboxymethyl chitosan film solution and starch film solution were used to coat pomelo fruit(Guanxi Honey Pomelo).The coated fruit were storaged at room temperature,and their physiological and biochemical parameters were determined,such as pulp organic acids,respiration rate,ethanol concentration,weight loss,catalase activity,etc.The results showed that the coated fruit were lower in all the parameters determined than the control,and that the coatings effectively extended the shelf life of the pomelo fruit.The pomelo fruit had a shelf life of upto 80 days when coated with starch film solution,upto 90 days when coated with the carboxymethyl chitosan film solution,and upto 120 days when coated with the konjac glucomannan complex film solution.
Effect of NaCl Treatment on Na+ and K+ Content in the Leaves of Sesuvium portulacastrum

Abstract:
Although regarded as one of the major cations contributing to soil salinity,sodium was also reported to be beneficial to plant growth.To elucidate the function of sodium in plants under salt stress,seedlings of Sesuvium portulacastrum were treated with NaCl at different concentrations(0,300,600,900 mmol.L-1) to observe the leaf morphology,biomass weight and Na+ and K+ content.The results showed that the seedlings treated with 300 mmol.L-1 NaCl produced the highest biomass.The flame emission and X-Ray microanalysis showed that large amounts of sodium ions were accumulated in the leaves,and that the leaf Na+ content was increased dramatically with the NaCl concentration while the leaf K+ content decreased,which leads to a high Na+ /K+ ratio.This study suggests that sodium of a given range of concentration could improve plant growth but high levels of sodium reduce uptake of K+ by the plant,hence leading to toxicity to the plant growth.

Effects of Temperature and Salinity on Larval Development of Anadara antiquate

Abstract:
Responses of Anadara antiquate larval to various ambient water temperatures and salinity levels were measured in terms of hatchability and larval development rate.The results showed that A.antiquate developed to D-model larvae from fertilized eggs for about 22 hours and 45 minutes when incubated at the ambient temperature of 28 ℃ and the salinity level of 28.The optimum ambient temperature for larval development ranged between 25~29℃,within which A.antiquata had a higher embryonic development rate and a higher hatchability but a low malformation rate.The optimum ambient salinity for larval development was 22~30,at which A.antiquata gave a higher hatchability and a higher larval development rate,and the larvae hatched from the eggs grew well.

Characteristics of the Phytoplankton Community in Xincun Harbour,Lingshui,Hainan

Abstract:
A red tide survey of 14 voyages was made in the Red Tide-Monitoring Areas of Xincun Harbour in Lingshui,Hainan during the period from April to October 2011,and recorded therein were phytoplankton species composition,dominant species,density,Shanon-Wiener index(H’) and other community characteristics,based on which the horizontal distribution and seasonal variation trend of the phytoplankton community were analyzed.The results showed that a total of 106 species were recorded,of which diatom was dominant.However,the dominant species changed with time.The number of phytoplankton individuals was significantly different in horizontal distribution,was higher inside the harbour than near the entrance,and was remarkably affected by temperature.

Abstract:
A survey of aquatic plants used for landscaping was made in the residential areas of Haikou City,Hainan,China,and registered 29 species of aquatic plants under 26 genera and 18 families.This survey presents problems on use of aquatic plants for waterscaping in the residential areas,such as low species diversity,unreasonable plant selection and arrangement,poor waterscaping ecological effect,poor maintenance and care of the aquatic plants in waterscaping,etc.Some suggestions on solution to the problems are raised herein.

Abstract:
Late embryogenesis abundant proteins(LEA proteins) are a group of stress-responsive proteins in higher plants that are induced by environmental stress.These proteins can enhance plant tolerance against environmental stress via stabilizing cell membrane and binding ions.The category and structure of LEA proteins are summarized herein.The researches conducted in recent years are reviewed about the roles of LEA proteins in plant response to drought,low temperature and other environmental stresses.
