Abstract:
Based on SGS conservative amino acid region of Sgt1 gene from different crops,degenerate primers were designed and RT-PCR was used to amplify SoSgt1 cDNA fragment of chewing cane. The results indicated thatSoSgt1 full length cDNA from chewing cane includes 1438 bp,362 amino acids. Protein Blast from NCBI database was used to analysis its amino acid sequence,and the results showed that the SoSgt1 protein have three SGT1 typical conserved domains eg. TPR,CS and SGS,and the similarity among SGT1 of other crops were very high. The expression levels ofSoSgt1 increased after inoculating with Gibberella fujikuroi. Based on SoSgt1 cDNA ,two restriction sites were designed and antisense expression vector pCAM-SGT were constructed,in which plant eukaryotic expression vector,pCAMBIA1301,were used as backbone.

AtNUDT8 was cloned by PCR from Arabidopsis thaliana and inserted into pCHF3 to form a recombined expressing vector,pCHFN8. Agrobacterium tumefaciens mediating method was used to transform this vector into wide type A. thaliana. And a batch of transgenic A. thaliana over-expressing AtNUDT8 was obtained through kanamycin screening and confirmed by Northern blot.

Abstract:
In order to extract high quality protein sample for two dimensional electrophoresis (2-DE) assay,aqueous solution method,trichloroacetic acid-aceton method and the advanced phenol isolated method were used to extract the proteins from Rhizophora apiculata Bl,and the harvest rate,the integrality and the solubility of the three methods were compared and analyzed. The results showed that the colloidal deposition,which was composed of polysaccharide but little of protein,was isolated by the aqueous solution method; the proteins,which were isolated by trichloroacetic acid-aceton method,was purer than that from others and the background in 2-DE was most clearest,but the integrality of protein sample was not enough for 2-DE; 800—1000 μg protein deposition could be harvested from 500 mg leaves material by the advanced phenol isolated method,and the protein spots which were detected on 2-DE gel map were seven times than that by trichloroacetic acid-aceton method. These data demonstrated that the advanced phenol extraction method was more excellent than others based on the great protein harvest ratio and integrality.

Abstract:
5% vanillin acetum and perchloric acid were used as the color developer,and the pure ursolic acid was used as the reference,the contents of total triterpenoid in adult leaves of 6 Kudingcha species in Aquifoliaceae and 4 kind marketed shoots products of Ilex kudingcha C. J. Tseng were determined by spectrophotometry at 548 nm. The results showed that the adult leaves of I. latifolia Thunb. had the highest total triterpenoid,and the marketed shoots products of I. kudingcha C. J. Tseng had the lowest total triterpenoid on average. The contents of total triterpenoid in 6 Kudingcha species in Aquifoliaceae for test samples were sorted as follows:I. latifolia Thunb. (4.209%)>I. cornuta Lindl. (3.866%)> I. huoshanesis Y. H. He (3.064)> I. centrochinesis X. Y. Hu (2.775%)> I. kudingcha C. J. Tseng (2.74%)>I. pentagona S. K. Chen,Y. X. Feng et C. F. Liang (2.648%).

Abstract:
Transmission electron microscopy was used to observe the ultra-structure of spermatid of Trochus pyramis Born at different stages. The results indicated that the spermatogenesis of Trochus pyramis Born included following five stages:spermatogonium,primary spermatocyte,secondary spermatocyte,spermatid and spermatozoon. The shape of spermatogonium was ellipsoidal and there were few mitochondria,the distribution of chromatin uniform; the shape of primary spermatocyt was much bigger,the chromatin agglomerated as pieces and there were more mitochondria; Compared with primary spermatocyte,secondary spermatocyte was smaller and there were more mitochondria,and some mitochondria fused and taken on cisterna shape. Spermiogenesis could be divided into five stages,the characteristics of the first stage:electron dense granules participated in the formation of acrosome; that of the second stage:the nucleus presented ellipsoidal shape or spherical shape,and become long cylinder gradually; that of the third stage:the chromatin agglomerated as larger pieces; that of the fourth stage:the mitochondria fused gradually,the volume improved and cristae developed; that of the fifth stage:centrosome moved and flagellar formed,the chromatin agglomerated further and become higher electron-density homegeneous,cytoplasm diminished. Mature spermatozoon was primitive type and composed of head,middle piece and tail.

Abstract:
To analysis the reasons why spiraling whitefly move,based on the known biological characteristics and partial activity patterns,artificial climate chamber and some other equipments were used to study the effects of temperature and light change on take-off activity of spiraling whitefly Aleurodicus disperses. The results showed that the flying number began to increase since 15 ℃,the optimum taking off temperature was 30 ℃. From darkness to light,more whiteflies took off. With the change of light,male whiteflies made courtship response. The sex ratio of Aleurodicus disperses taking off was above 1.

In the paper,the constructed mathematical models,together with choose assessment method,were used to analysis the main source of measurement uncertainty of Pb and Cd measured by ICP-MS,after analyzing and synthesizing of six components,the samples weighing,digestion,the volume size,determination of the standard process of preparation,curve fitting,equipment measurement repeatability in the process of anglicizing,the expanded uncertainty of Pb and Cd was calculated. These data provided references for the reliability research of the accurate extent and method for analyzing measure results.

Our research aims to find an agricultural intercropping eucalyptus model,which promotes the sustainable production of eucalyptus forest,and to some extent,alleviate the shortage of land resources in China. The analysis data showed that east-west line big wide mode of eucalyptus agricultural intercropping system was as far as possible to reduce the degree of shade,and by which the sun light could enter the intercropping with crops to the greatest extent,and satisfied the need for light and heat from the crops.

Abstract:
The mycelium growth rate method was used to measure and compare the toxicities of 5 fungicides against R-HKB and R-DZC,of Rhizoctnia solani Kühn,which were 25% propiconazole,12.5% tebuconazole,80% ethylicin,25% prochloraz and 5% Jinggangmycin,The results showed that propiconazole was the most effective on inhibiting the radial growth of strain R-HKB mycelium,and its EC50 was 0.013 2mg·L-1;tebuconazole was the most effective on inhibiting the radial growth of R-DZC mycelium,and its EC50 was 0.413 1mg·L-1;The toxicity of Jinggangmycin was the lowest against the growth of R-HKB and R-DZC mycelium,whose EC50 were 714.494 8 mg·L-1 and 663.417 1 mg·L-1 respectively.The toxicity of Jinggangmycin was 54 128.4 and 1 606.0 times less than that of propiconazole and tebuconazole separately.These data indicated that propiconazole and tebuconazole were more effective for the growth inhibition of R.solani in Aquilaria sinensis and Zoysiagrass sinica.

Abstract:
During somatic embryogenesis of Cassava[Manihot esculenta Crantz(M. utilissima Pohl)],the activities of SOD (superoxidase) decreased when the embryogenic cells were developing to the globular embryos,while increased when the globular embryos were developing to the cotyledon embryos. However,the activities of POD (peroxidase) and CAT ( catalase) showed a reverse trend to that of SOD. It suggested that the decreased SOD activity might be an index of the differentiation of embryogenic cells and early development of embryos in Cassava. During the differentiation and mature of embryogenic cells,the isozyme spectra of POD increased from five to nine,and five of them were stable.

The coding regions of papaya eIF4E and eIFiso4E were amplified by RT-PCR and then fused with pBD-GAL4 vector. After confirmation with sequence analysis,the plasmid was transformed into the yeast cell,YRG-2,and the toxicity and transcriptional autoactivation of expressed protein were tested. The results indicated that the coding regions were successfully amplified and subcloned into pBD-GAL4,and the YRG-2 transformed with bait plasmids grew well on SD/trp plate but not on SD/-his-trp. These data showed that the bait vectors,pBD-GAL4-eIF4E and pBD-GAL4-eIFiso4E,were expressed correctly without toxicity,and could not autoactivate the transcription of reporter gene alone in yeast two hybrid systems,and could be used for analyzing the interaction between papaya eIF4E,eIFiso4E protein and virus.

L-phenylalanine N-hydroxylase is the first enzyme which catalyze the biosynthesis of benzylisothiocyanate (BITC) in plant. According to the sequences of CYP79A2 gene of Arabidopsis thaliana (NM120608) and Brassica rapa(EU877074)from GenBank and combining with the genome draft of the transgenic tropical fruit tree papaya,specific primers were designed and RT-PCR was used to amplify partial sequences of CYP79A2 gene from papaya young leaves. The sequence includes 923bp,which named CPCYP79A2. Bioinformatics analysis results showed that the frgment is the partial sequences of CYP79A2 gene of papaya. The data set a strong basis for the study of the biosynthetic regulation of BITC in papaya.

In order to guarantee the smooth development of banana growing,from April,2004 to May,2006,the diseases of banana crop from the main banana producing areas in Hainan Province,including Danzhou,Baisha,Changjiang,Dongfang,Ledong,and Sanya,were investigated and identified. Twenty-seven kinds of diseases on bananas were found,18 of which belong to fungal,3 belong to bacterial,4 belong to nematodes and 2 belong to viruses. The following 7 kind major diseases,banana wilt,anthracnose,macrophoma spot,bunchy top,cordana leaf spot,alternaria leaf spot and heart rot mosaic,damaged banana crop in Hainan.

Based on the information of EST sequences from latex SSH cDNA library stimulated by ethephon,RACE was used to isolate the full length cDNA of an unknown functional novel gene with two LIM domains from H. brasiliensis,HbLIM. Bioinformatics analysis results showed which comprises of 1 016bp with a 570bp ORF and encodes a 189 animo acid residues. The deduced animo acid sequence with two LIM domains and two N-myristoylation sites is rich in Lys,Ser,Ala and Leu and has high identity of 98%,95% and 92% to the homologues from Pollen specific,Populus trichocarpa and Vitis vinifera. These results were very important for elucidation of the functions of LIM-domain protein in latercifer development and metabolism in H. brasiliensis.

Carcass traits and meat quality traits of Hainan Pigs,Black Pigs ( Hainan Pigs♀×Duroc♂) were measured,and the genetic variation of H-FABP and MyoG 5’ upstream sequence of Hainan Pigs were analyzed. The results indicated that the live weight,carcass weight,dressing percentage,carcass length and lean yield of Black Pigs were higher than that of Hainan Pigs (P < 0.05),bone percentages,skin percentages were lower than that of Hainan Pigs,fat deposition capability was higher than that of Hainan Pigs,and the changes of loin eye area and skin thickness were not significant. The meat quality of Hainan Pigs and Black Pigs attained fine meat quality criteria,drip loss,marbling,moisture,crude protein concentration of Black Pigs were superior to that of Hainan Pigs. The gentype and gene frequency of genetic variation of H-FABP and MyoG 5’ upstream sequence of Hainan Pigs set an important genetic basis for the formation of fine meat quality traits of Hainan Pigs.

Abstract:
Three mulitivariation analysis methods (cluster analysis,principal component analysis and discriminate analysis) were used to compare the morphological characters of four populations of Paphia euglypta in Yangjiang,Zhanjiang,Haikou and Beihai. The results of clusters analysis and principal component analysis indicated that the morphological characters were similar among the populations in Yangjiang,Zhanjiang and Haikou,but quite different from that of Beihai. In the principal component analysis,two principal components were constructed,the contributory ratio of the first principal component was 32.840%,that of the second principal component was 26.106%,and the cumulative contributory ratio was 58.945%. The results of discriminant analysis revealed that there were significant differences among the four populations (P < 0.01). The identification accuracy was 46.7%~100% (P1) and 40.5%~96.7% (P2). The total discriminant accuracy was 64.7%. The Mantel test results showed that Euclidean distance was positively correlated with geographical distance (r=0.0974,P=0.5770).

Conotoxins (CTXs) are one kind active peptide from venom ducts of predatory snails of the genus Conus,which possess low molecular mass,novel structure and high selectivity for different receptors’ subtypes. CTXs can be used as candidates for novel drugs or potential drug leads. Based on ligand binding sites,conus peptides were divided into different classes.α-CTXs are one of important kind CTXs which share the following arrangement of cysteine residues in the primary amino acid sequence:CC-C-C. According to the number of amino acids between cysteine residues,α-CTXs were further divided into different subfamilies. α-CTXs are selective antagonists of neuromuscular or neuronal nicotinic acetylcholine receptors (nAChRs),which are very important in clinic therapy for neuralgia,Parkinson’s disease,dementia ,Alzheimer’s disease and small cell lung cancer et al. In the paper,the structure,classification and the interaction of α-CTXs and nAChRs were reviewed.

Abstract:
Duration of latex flow after tapping is a crutial factor that limits rubber yield,which is mainly determined by the plugging formation at the end of severed laticifers. The protein in latex plays an important role in the regulation of rubber particle aggregation,which is closely associated with the plugging formation. In order to provide clues for elucidating the mechanism by which the severed laticifers are plugged and for exploring molecular markers applied in predicting the yield potential and ability of latex flow of clones or wild germplasms of rubber tree in Hevea breeding program,the progresses of the isolation and characterization of proteins related to rubber particle aggregation in latex were reviewed.
