Effects of Some Ecological Factors on the Absorption of N ,P by Enteromorpha linza Ulva

Abstract:
To explore the feasibility of Enteromorpha linza ulva in optimizing shrimp culture environment,the effects of light,pH,salinity,temperature and other ecological factors on the absorption of N,P by Enteromorpha linza ulva were analyzed. The results indicated that N,P by Light,pH,salinity,temperature and other ecological factors affected the uptake rate N,P by Enteromorpha linza ulva significantly. Within 0—1 000 lx light intensity,the uptake rate of DIN,DIP by Enteromorpha linza ulva increased with the increase of light intensity; within 1 000—5 000 lx light intensity range,the uptake rate of DIN,DIP were higher,average was(0.000 30±0.001 2)μmol·g-1·h-1,(0.001 56±0.001 1)μmol·g-1·h-1; above than 5 000 lx,the uptake rate of DIN,DIP decreased significantly. Within pH 7.0—9.0,the uptake rate of DIN,DIP were higher,average was(0.000 12±0.001 2)μmol·g-1·h-1,(0.001 68±0.001 42)μmol·g-1·h-1,while outside this range,the uptake rate of DIN,DIP decreased significantly; Within the salinity range of 15—25,the uptake rate of DIN,DIP were higher,the highest vlue was observed when salinity in 15,the uptake rate of DIN,DIP were(0.000 21±0.000 05)μmol·g-1·h-1 and(0.005 5±0.000 6)mol·g-1·h-1. Within 20—35℃,the uptake rate of DIN,DIP were higher,the maximum absorption rate of P reached at 35℃,which was (0.000 56±0.002 6)μmol·g-1·h-1,the maximum absorption rate of N maximum reached at 30℃,which was (0.024 56±0.000 11)μmol·g-1·h-1,while outside the scope ,the absorption rate decreased significantly.

Abstract:
The method of improved CTAB,SDS and QIAGEN RNeasy Plant Mini kit were used to isolate total RNA from juvenile and senescence banana roots respectively,and the quality,quantity and purity of total RNA were compared. The results showed that the quality,quantity and purity of total RNA isolated by improved SDS were superior to that by other methods.

Abstract:
In our report,rice variety Nipponbare genome were used as template,a pair of primers which were added a XcmⅠ site at each 5’ end were used to amplify,and a 627bp fragments were obtained,which was ligated into pGEM-T Easy to develop a recombinant vector,T-xia. Self-made T vector was obtained when T-xia was digested by XcmⅠ,and the results of the ligation with other PCR products indicated that 80% recombination clones contained target fragments.

Abstract:
After selection,13 strain Bacillus licheniformis with high protease activity were obtained from the sea water and sediments of the aquaculture systems and the natural sea area in Wenchang and Haikou,Hainan province,which were identified to be the wild tropic strains of Bacillus licheniformis by morphological analysis,physiological and biochemical analysis and 16S rDNA sequencing.
Isolation of the Protoplast from Dendrobium Nobile Lind1 PLBs

Abstract:
In order to obtain regeneration Dendrobium nobile Lind1 through protoplast isolation and cultivation,protoplasts were isolated from PLBs of Dendrobium nobile Lind1. The effects of various factors (composition of enzymes,digestion time,concentrations of osmotic pressure stabilize (mannitol),centrifugation speed,purification methods,pretreatment of the material and the methods of vigor test) on the yield,viability and division of protoplasts were investigated. The results indicated that the optimum condition for protoplast isolation was established by using 1% cellulase R-10 and 0.4% macerozyme R-10 in 0.65 mol·L-1 mannitol (pH5.7) solution for cell wall digestion; the mixture of PLBs and enzyme solution was incubated at (25±2)℃ in dark for 6h on a rotary shaker with an agitation speed of 60 rpm,with adoption of 50—100μm filters to remove undigested tissues and debris,and after centrifugated at 800 rpm for 4min,the maximum yield (8.25±0.17)×105 per gram PLBs and the high viability (90.24±1.84)% were obtained. These data suggested that the small meristem cells of PLBs were good source of intact protoplast.

Abstract:
The effects of temperature,illumination intensity,salinity and pH on the growth and reproduction of Melosira sp. were analyzed under the experimental conditions. The results indicated that the adaptive water temperature ranged from 5 ℃ to 30 ℃ with the optimal temperature at 25 ℃; the adaptive illumination intensity was 500—9 000 lx with optimal value at 3 000—7 000 lx; the range of adaptive salinity was 5—85 with the optimal salinity at 20; the range of adaptive pH was 4—10 with the optimal pH at 8.5.
Optimization on the Preparation of the Competent Cells of Escherichia coli HB101

Abstract:
The strain source,the growth phrase of the cell and the storage condition were optimized for preparing the competent cells of Escherichia coli HB101. The results revealed that the highest transformation ability could be obtained when the source of the strain was streaked from strains stored under-70 ℃,cells were harvested when OD600 were 0.3—0.4,the storage condition of competent cells was under 4 ℃ for 1 d and-70 ℃ for 1 month,and the highest transformation efficiency attained 1.18×107 cfu·g-1 DNA.
Resistance Risk Assessment and Cross-Resistance of Plutella xylostella to Abamectin

Abstract:
A laboratory selection was conducted for Plutella xylostella resistance to abamectin. The results showed that the resistance developed slowly at first and then quickly during the selection. After 14 times of selection during the 20 generations,P.xylostella exhibited 20.92-folds resistance to abamectin compared to the unselcetde parent strain. Estimation of realized heritability (h2) of resistance was studied based on the Tabashnik’s method and the h2 for the entire selection experiment was 0.1303. It required 2.4 to 15.9 generations for P.xylostella to obtain 10 fold increase in resistance ratio under selective pressure of 50% to 99% mortality for each selective generation. These results suggested that P.xylostella might have the risk to develop high resistance to abamectin. The susceptibility of the resistant strain to other five insecticides was also tested. The results indicated that the resistant strain had an obvious cross-resistance to emamectin benzoate with a resistant ratio was 8.85 fold,but it had no or low cross-resistance to spinosad,indoxacarb,fiprronil and chlorfenapyr.
Introduction Experiments of Anti-season Eggplant in Hainan

Abstract:
In order to solve the problems of off-season cultivation of eggplant in Hainan,such as commodity decline,production instability,disease resistance and so on,11 eggplant varieties were introduced and tested. The results showed Changfeng No 2 was precocity,high resistance to powdery mildew,and had 15.6% higher output more than that of Suike No 2,which was the main cultivation varieties in Hainan province,and that the characteristics of the long rod type fruits,dark purple epidermis and high commodity rates conformed to the demand of the consumer in Hainan province and adjacent areas.
Effects of Temperature on Content of Four Materials in Leaves during Gender Transition of Hermaphrodite Papaya(Carica papaya L.)

Abstract:
In the paper,the content changes of protein,starch,moisture and dissociated amino acid in the hermaphrodite plant under different high temperature were analyzed. The results showed that the protein content in leaves decreased with temperature increasing,which decreased 28% at 38 ℃ than that at 33 ℃,and decreased 1.3% at 41 ℃ than that at 38 ℃; the starch content in leaves decreased with temperature increasing too,which decreased 2.9% at 35 ℃ than that at 33 ℃,and decreased 20% at 38 ℃ than that at 35 ℃; with temperature increasing,the moisture content in leaves decreased firstly and then increased,which decreased 1.9% at 35 ℃ than that at 33 ℃,but increased 1.4% at 38 ℃ than that at 35 ℃; the content of dissociated amino acid in leaves between 33 ℃ and 34 ℃,at which the gender of hermaphrodite were stable,those between 37 ℃ and 38 ℃,at which the gender of hermaphrodite were changed,but some of which ,such as aspartic acid,glycin,arginine and proline increased. These data indicated that the gender transition of hermaphrodite papaya might be associated with the content changes of protein,starch,moisture and dissociated amino acid in the hermaphrodite plant under different high temperatures.
Investigation of Harmful Mollusks in Facility Cultivation of Tropical Orchids in Hainan

Abstract:
The destruction of the mollusks has become one of the most important factors to restrict the tropical orchid production. In our report,the harmful mollusks in the orchid gardens in the northern regions of Hainan Island (Haikou),central regions of Hainan Island(Qiongzhong)and southern regions of Hainan Island(Sanya) were investigated. The results indicated that 5 species of snails,which were Achatina fulica (Bowdich,1822),Camena platydons( Benson,1851),Acusta tourannensis rhodostoma (Moellendorff,1884),Succinea orientalis (Pfeiffer,1845) and Microcystina sinica (Moellendorff,1885),and 2 slug species,one of which was Meghimatium bilineatum(Benson,1842)and another one which was not identified was extremely rare in Hainan,were detected. Based on these data,the harm extents of them were analyzed and some prevention and elimination suggestions were proposed.
Effects of 1-MCP on Some Physiological Metabolic Indexes of Curuma alismatifolia Cut Flowers During Storing

Abstract:
Physiological changes of Curcuma alismatifolia cut flowers during storing,as well as the effects of fumigation method for 1-MCP on the physiological indexes and on the metabolism of active oxygen species were investigated. The results showed that,with the extension of the shipping time,the weightiness rate of the cut flower increased significantly,petal membrane permeability and anthocyanin increased significantly,while ethylene production decreased in general,during shipping and storing,different degree concentration deals inhibited the incresement of four physiological indexes in various degree; in the later period of shipping and storing,certain concentration of suffocating inhibited the augment of petal membrane permeability and releasing of ethylene observably,but had no significant effect on the increasement rate of weightiness; on the day five,four different degree concentration deals all inhibited SOD and PODs’ activities,the produce of MDA was inhibited by 2-4 μL·L-1 of 1-MCP significantly,except for O-2.4 μL·L-1 of 1-MCP had the best effect on active oxygen metabolizing. So,4 μL·L-1 1-MCP was the best dosage for the storing of curcuma alismatifolia cut flowers.
