Cloning of Na+/H+ Antiporter Gene from Bruguiera gymnorrhiza(L.) LAM

Abstract: According to the homologous sequences from Atriplex gmelini,Populus euphratica,Tetragonia tetragonioides,Kalidium foliatum,Populus tomentosa and Salicornia europ,PCR primers were designed and RT-PCR and RACE were used to amplify NHX1 DNA fragment from the leaves of Bruguiera gymnorrhiza.The results indicated that the BgNHX1 contains 2 130 bp,including an open reading frame of 1 626 bp,which encodes a predicted polypeptide of 541 amino acids,and there are 12 predicted transmembrane domains.Base on the data that the sequence showed high homologies(> 80%) with Na + /H + antiporter genes from Ricinus communis Populus euphratica,Citrus,paradisi,vitis vinifera,it was very possible that BgNHX1 was competent for Na + compartmentation in cells.
